目的建立超高效液相色谱法(UPLC)同时测定阿托伐他汀钙原料药中15个杂质含量的方法。方法采用岛津Shim-pack Velox PFPP(2.1 mm×100 mm,1.8μm)色谱柱,以3.9 g/L醋酸铵缓冲液(pH 5.0):乙腈:甲醇:四氢呋喃(无稳定剂)=67:21:6:6为流动相A,3.9 g/L醋酸铵缓冲液(pH 5.0):乙腈:甲醇:四氢呋喃(无稳定剂)=27:61:6:6为流动相B;梯度洗脱,流速0.43 ml/min,检测波长244 nm,柱温35℃,样品室温度10℃;进样量1.8μl。结果阿托伐他汀钙及各杂质峰间分离良好,在0.3~3μg/ml范围内15个杂质均呈良好的线性关系(r均>0.999,n=6),各杂质平均回收率分别为96.3%,99.1%,99.9%,102.4%,96.4%,99.8%,99.4%,104.9%,106.4%,105.4%,100.1%,98.9%,94.7%,94.4%,101.4%,且不同浓度间平行性良好。15个杂质重复性、进样精密度、中间精密度均良好,48 h内均较稳定。此法除对色谱柱要求较高外,耐用性良好。结论本方法简便、快速、分离度好、专属性强,经方法学验证可用于阿托伐他汀钙原料药杂质质量控制。  相似文献

Magnesium, as a biodegradable metal, offers great potential for use as a temporary implant material, which dissolves in the course of bone tissue healing. It can sufficiently support the bone and promote the bone healing process. However, the corrosion resistance of magnesium implants must be enhanced before its application in clinical practice. A promising approach of enhancing the corrosion resistance is deposition of bioactive coating, which can reduce the corrosion rate of the implants and promote bone healing. Therefore, a well-designed substrate-coating system allowing a good control of the degradation behavior is highly desirable for tailored implants for specific groups of patients with particular needs. In this contribution, the influence of coating formation conditions on the characteristics of potentiostatically electrodeposited CaP coatings on magnesium substrate was evaluated. Results showed that potential variation led to formation of coatings with the same chemical composition, but very different morphologies. Parameters that mostly influence the coating performance, such as the thickness, uniformity, deposits size, and orientation, varied from produced coating to coating. These characteristics of CaP coatings on magnesium were controlled by coating formation potential, and it was demonstrated that the electrodeposition could be a promising coating technique for production of tailored magnesium-CaP implants.  相似文献

PurposeTo gain a better understanding of eye care professionals’ (ECPs) perceptions regarding the benefits of silicone hydrogel (SiH) daily disposable contact lenses (DDCL), particularly with respect to health, comfort and patient satisfaction.MethodsA survey was conducted with 300 ECPs in the United States, United Kingdom and Japan during November 2017. The survey comprised 34 statements relating to SiH DDCLs, to which the ECPs provided their level of agreement using a 6 point Likert scale. A minimum of 70% agreement was set to define majority agreement. Categories of statements included Health, Comfort, Patient Experience, and Standard of Care.ResultsECPs rated the highest levels of agreement to perceptions within the Patient Experiences and Health categories. The six statements receiving the highest ratings were “Silicone hydrogel 1 day lenses satisfy today’s patients’ demanding lifestyles” (93% agreement); “Silicone hydrogel 1 day lenses are the best choice to safeguard my patients’ eye health related to contact lens wear” (92%); “Silicone hydrogel 1 day lenses provide the best benefits to my patients” (92%); “Silicone hydrogel 1 day lenses provide better long term eye health for my patients than hydrogel 1 day lenses” (91%); “Silicone hydrogel is the healthiest lens material for my daily disposable patients” (90%); and “Silicone hydrogel 1 day lenses provide a better wearing experience for my patients than hydrogel 1 day lenses” (90%).ConclusionECPs perceive that SiH DDCLs offer long-term eye health and comfort for the patient and although the ECPs surveyed would not necessarily prescribe SiH DDCLs to all their patients, they would prescribe them to most of their patients. The results support the premise that while ECPs consider SiH 1 day contact lenses as the current “standard of care”; the principal barrier continues to be the perceived higher cost of these lenses.  相似文献

We describe the synthesis, characterization and direct‐write 3D printing of triblock copolymer hydrogels that have a tunable response to temperature and shear stress. In aqueous solutions, these polymers utilize the temperature‐dependent self‐association of poly(alkyl glycidyl ether) ‘A’ blocks and a central poly(ethylene oxide) segment to create a physically crosslinked three‐dimensional network. The temperature response of these hydrogels was dependent upon composition, chain length and concentration of the ‘A’ block in the copolymer. Rheological experiments confirmed the existence of sol–gel transitions and the shear‐thinning behavior of the hydrogels. The temperature‐ and shear‐responsive properties enabled direct‐write 3D printing of complex objects with high fidelity. Hydrogel cytocompatibility was also confirmed by incorporating HeLa cells into select hydrogels resulting in high viabilities over 24 h. The tunable temperature response and innate shear‐thinning properties of these hydrogels, coupled with encouraging cell viability results, present an attractive opportunity for additive manufacturing and tissue engineering applications. © 2018 Society of Chemical Industry  相似文献

对锂辉石-氧化钙烧结过程进行热力学分析,绘制了各反应Gibbs自由能与温度的关系图。结果表明,Al2O3会优先和Na2O、Li2O、K2O反应,然后与CaO反应生成CaO·Al2O3,而且烧结温度需高于1060℃以保证LiAlSi2O6能够完成晶形转变。并探讨了锂辉石-氧化钙烧结法提锂的反应机理。考察了不同烧结条件对锂浸出率的影响并对熟料进行X射线衍射(XRD)分析表征。实验结果表明,在配料比为1∶1.25、烧结温度1150℃、烧结时间60min时,锂的浸出率达到92.14%,熟料中的主要物相为Ca2SiO4与LiAlO2。利用XRD和扫描电镜-能谱联用仪(SEM-EDS)对熟料与浸出渣的物相、显微形貌及元素分布情况进行了分析表征。为了确定烧结反应的控制性步骤,在最优烧结条件的基础上对烧结过程进行动力学分析,结果表明,锂辉石-氧化钙烧结体系属于球形颗粒三维界面化学反应控制,烧结过程的动力学拟合方程为1-(1-x)1/3=0.00677t。  相似文献

New treatment strategies for spinal cord injury with good therapeutic efficacy are actively pursued. Here, acetalated dextran (AcDX), a biodegradable polymer obtained by modifying vicinal diols of dextran, is demonstrated to protect the traumatically injured spinal cord. To facilitate its administration, AcDX is formulated into microspheres (≈7.2 µm in diameter) by the droplet microfluidic technique. Intrathecally injected AcDX microspheres effectively reduce the traumatic lesion volume and inflammatory response in the injured spinal cord, protect the spinal cord neurons from apoptosis, and ultimately, recover the locomotor function of injured rats. The neuroprotective feature of AcDX microspheres is achieved by sequestering glutamate and calcium ions in cerebrospinal fluid. The scavenging of glutamate and calcium ion reduces the influx of calcium ions into neurons and inhibits the formation of reactive oxygen species. Consequently, AcDX microspheres attenuate the expression of proapoptotic proteins, Calpain, and Bax, and enhance the expression of antiapoptotic protein Bcl‐2. Overall, AcDX microspheres protect traumatically injured spinal cord by alleviating the glutamate‐induced excitotoxicity. This study opens an exciting perspective toward the application of neuroprotective AcDX for the treatment of severe neurological diseases.  相似文献

基质囊泡(Matrix vesicles,MVs)是骨矿物初始形核生长的场所。骨矿化过程中,钙、磷酸根等离子经通道蛋白跨膜运输进入MVs内,当局部浓度达到一定值时,磷酸钙晶体开始沉积。磷酸钙的存在形态包括无定形磷酸钙、磷酸八钙及羟基磷灰石等。MVs可以调节细胞内外基质中的钙和磷酸根离子的稳态及无机磷酸盐/无机焦磷酸盐的比值,提供磷酸钙晶体成核位点,在骨矿化的初始启动过程中发挥重要作用。本文概述了MVs的生物来源、分子组分、提取方法,MVs介导的骨矿化过程,以及近年来利用囊泡作为体外矿化模型模拟MVs矿化过程的研究进展。  相似文献

Mechanically exfoliated 2D hexagonal boron nitride (h-BN) is currently the preferred dielectric material to interact with graphene and 2D transition metal dichalcogenides in nanoelectronic devices, as they form a clean van der Waals interface. However, h-BN has a low dielectric constant (≈3.9), which in ultrascaled devices results in high leakage current and premature dielectric breakdown. Furthermore, the synthesis of h-BN using scalable methods, such as chemical vapor deposition, requires very high temperatures (>900 °C) , and the resulting h-BN stacks contain abundant few-atoms-wide amorphous regions that decrease its homogeneity and dielectric strength. Here it is shown that ultrathin calcium fluoride (CaF2) ionic crystals could be an excellent solution to mitigate these problems. By applying >3000 ramped voltage stresses and several current maps at different locations of the samples via conductive atomic force microscopy, it is statistically demonstrated that ultrathin CaF2 shows much better dielectric performance (i.e., homogeneity, leakage current, and dielectric strength) than SiO2, TiO2, and h-BN. The main reason behind this behavior is that the cubic crystalline structure of CaF2 is continuous and free of defects over large regions, which prevents the formation of electrically weak spots.  相似文献

高倍率灰钙循环烟气脱硫技术在神东矿区的煤粉锅炉环保改造项目中已投入建设并成功运行,脱硫率可达95%以上。在NGD系统的运行调试期,布袋除尘器压差的数值及其上升速度均较改造前有大幅提高,影响了锅炉系统的正常运行并降低了布袋除尘器的运行效率。为了解决此问题,先分析滤料透气性降低而导致布袋除尘器压差上升的根本原因,再结合现场调研情况对NGD系统工况下的烟气含湿量、烟气温度、烟气灰浓度、除尘器本体的问题以及上游锅炉系统问题逐一进行对比和分析,并找出问题产生的各主要因素。NGD系统中布袋除尘器差压升高的原因包括多方面,为了使NGD系统的运行更加稳定,需要使除尘器适应NGD系统的工况并对系统中部分设备进行改造。在未来NGD系统的设计中,布袋除尘器需按高浓度烟气设计,且针对煤粉工业锅炉的运行、维护特性应加入对烟气中含水量的监测及对布袋除尘器的相应保护措施。  相似文献
